Osmoregulation: Cyclic GMP and the big squeeze

چکیده

The second messenger cyclic GMP (cGMP) is a biological enigma. Chemically, it is very similar to cyclic AMP, a signalling molecule whose functions are well understood and well conserved throughout eukaryotes. The role of cGMP is much less clear, however, and varies remarkably between cell types. A recent report [1] reveals a novel and unexpected function for cGMP, in promoting resistance to osmotic shock in Dictyostelium.

(Dys)regulation of epithelial chloride channels.

Chloride (Cl)-conducting channel proteins in plasma and intracellular membranes subserve a diversity of cellular functions, including neurotransmission, osmoregulation, pH regulation in organelles, acid secretion by parietal cells in the stomach, and salt absorption or secretion in exocrine glands (pancreas, sweat gland) and transport epithelia (trachea, intestine). With the notable exception o...

Inhibition by atrial and brain natriuretic peptides of endothelin-1 secretion after stimulation with angiotensin II and thrombin of cultured human endothelial cells.

We examined the inhibition by atrial natriuretic peptide (ANP) and brain natriuretic peptide (BNP) of endothelin-1 secretion stimulated by angiotensin II (ANGII) and thrombin using cultured human umbilical-vein endothelial cells. ANGII and thrombin dose-dependently stimulated immunoreactive (ir) endothelin-1 secretion. Human ANP(1-28) and human BNP-32 both inhibited such secretion in a dose-dep...

C-type natriuretic peptide inhibits thrombin- and angiotensin II-stimulated endothelin release via cyclic guanosine 3',5'-monophosphate.

We examined the inhibitory effect of porcine C-type natriuretic peptide (CNP) on endothelin-1 secretion stimulated by thrombin and angiotensin II (Ang II) in cultured porcine endothelial cells. The results were compared with the effects of atrial (ANP) and brain (BNP) natriuretic peptides. Thrombin and Ang II produced a concentration-dependent stimulation of immunoreactive endothelin-1 secretio...

A cyclic GMP-dependent signalling pathway regulates bacterial phytopathogenesis

Cyclic guanosine 3',5'-monophosphate (cyclic GMP) is a second messenger whose role in bacterial signalling is poorly understood. A genetic screen in the plant pathogen Xanthomonas campestris (Xcc) identified that XC_0250, which encodes a protein with a class III nucleotidyl cyclase domain, is required for cyclic GMP synthesis. Purified XC_0250 was active in cyclic GMP synthesis in vitro. The li...
